Balancing the burden of heterologous pathway expression with the need for robust cellular metabolism has always been an underlying principle for metabolic engineering projects. Here we present a cost and functionality-optimized pathway integration vector system for the cyanobacterial host, Synechococcus elongatus PCC 7942. This isocaudomer-based platform enables transcriptional and translational optimization of heterologous pathways in vivo. Part characterization and preliminary efforts towards a proof-of-principle alkaloid production study will be presented.
