Continuous flow crystallization is an attractive mode of operation, due to its ability to generate consistent product quality while requiring a smaller footprint and lower production costs than its batch counterpart. We present the development of an automated continuous crystallization platform at Pfizer as a part of the Flexible API Supply Technology (FAST) initiative. This lab-scale continuous crystallization consists of a three-stage continuous stirred tank reactor (CSTR) system. LabVIEWâ„¢ software was designed and written in-house for the purpose of automated analysis and to control various hardware including a flow sonicator, feed pumps, and temperature controllers. We demonstrate the integration of a variety of in-line process analytical technology (PAT) tools, including Focused Beam Reflectance Measurement (FBRM), EasyViewerâ„¢, Blaze, and Infrared (IR) spectroscopy within this environment. By integrating equipment, PAT, and control software into a single, automated platform, enhanced real-time process understanding, identification of optimal conditions, and acceleration of development timelines are realized.
