The Effect of Calcium Chloride in Modified CHO Cell Growth for Antibody Expression

The antibody is a protein molecule produced from CHO-cells as a secreted molecule and it is further purified to ensure the product quality attribute for the patients. During manufacturing of the protein, it has been shown that cell growth is a key factor to obtain the appropriate protein concentration of the molecule. Some cell growth discrepancies have been associated to specific nutrients within the CHO-cells culture media. Previous data indicates that the concentration of CaCl2 in the media may impact negatively CHO-cells growth. Therefore, this investigation was focused in measuring the growth of CHO-cells using cell culture media spiked with different CaCl2 concentrations. The experiment was performed in shake flasks at constant temperature, CO2 injection and base media content. The viable cell density, viability, pH, pO2, pCO2 and metabolites were measured for day 0 and day 3. After performed the experiment it was observed that the concentration CaCl2 affect cell in terms of the viable cell density (VCD). Statistical analysis shows that for 0M of CaCl2 less cell growth occurred, but for concentrations near 1M the cell growth was optimum.
