(224e) Investigation of CNT-Induced Bacteria Lysis and Protein Release

Carbon nanotubes (CNTs) were investigated for their potential use as lysis agents for bacterial cells. Recombinant Escherichia coli BL21 (DE3) expressing b-lactamase and Green Fluorescent Protein (GFP) were exposed to various concentrations of CNTs under various levels of agitation and exposure times. Fluorescence assay for GFP, enzymatic activity for b -lactamase activity, and Transmission Electron Microscopy (TEM) were used to characterize the extent of lysis. A correlation was also developed that related the extent of protein release to the concentration of CNTs and agitation rate. The correlation indicated that the agitation rate and CNT concentration affect lysis in a similar fashion.
