(124e) The Effect of Intra-Granular Binder Distribution On Single Granule Strength

Granule strength is important for further processing of granules in the pharmaceutical industry as it influences granule attrition rates and the strength of tablets that can be made from granules (Horisawa, Komura, Danjo, & Otsuka, 1995).  The current study evaluates the effect that the spatial distribution of binder solid bridges has on granule strength.  Granules are formed through single drop tests on a static powder bed.  These granule are excavated and dried at an elevated temperature.  Granule structure is measured through the use of x-ray microtomography for model systems in which x-ray attenuation coefficients can be used to distinguish primary particles from dried binder bridges.  This allows segmentation of the x-ray images and quantification of the spatial distribution of binder.  Once the binder distribution is quantified, granules are crushed to measure the breaking strength of the granules.  Implications on desired granule structure and process design are discussed.
